I'm going to talk about our paper in the New Journal of Physics
titled "Freeze-thaw cycles induce content exchange between cell size lipid vesicles". In this work
We wanted to find out more about how life could have emerged under prebiotically plausible conditions.
We were looking into some physicochemical factors that might explain how information between early protocells could have been exchanged.
Protocells are precursors of cells and here we use lipid vesicles as model protocells.
They're also called liposomes or GUV's which stands for giant unilamellar vesicles.
It is commonly believed that compartmentalization must have played an important role in the origin of life.
Compartmentalization is a key principle for life in general and it is likely that isolating chemical reactions in compartments was a
prerequisite for the emergence of life.
Another basic biological principle is exchange of information between
compartments or individuals in general as we also see in modern life for example in more complex organisms a sexual reproduction. Here
we wanted to investigate how protocells were able to exchange information, which means exchange some of their content.
They did not have a complicated protein machinery, like single-celled organisms like bacteria.
So the question is how could these simple precursors of cells that most likely did not contain any kind of protein have done that?
It is likely that they relied on physicochemical factors like environmental events to do so,
especially any kind of periodically reoccurring events. One parameter that generally oscillates over time is temperature.
It does so for example on a daily cycle.
Changes in temperature especially get interesting at around the freezing temperature of water.
Generally one scenario of where life could have emerged is in pools of water that periodically freeze and thaw.
Unrelated to compartmentalization and content exchange previous work has shown that in a system that undergoes freezing and thawing
certain chemical reactions important for the emergence of life are favored. In this work,
we look into physicochemical advantages of such an environment that could have enabled an exchange of content between early protocells.
So to look into content exchange between our model protocells, we generate two populations of lipid vesicles and
we freeze and thaw them.
What we see when we look at them after they fully thawed is that there is content mixing happening
And it is actually very thorough; but more about that later.
One important part of this work was to develop a high throughput method to study these vesicles under various environmental conditions and driving forces. In
future studies, we want to test a large range of different parameters and different prebiotically plausible molecules.
So to do so instead of commonly used containers like micro test tubes,
we use glass-bottom microtiter plates.
These allow high throughput, because we can compare many different conditions at the same time; one condition in each well of the plate.
But especially the glass bottom of these compartments plays an important role in our protocol, because it allows us to generate,
freeze and thaw and image our vesicles all in the same container without pipetting steps in between.
We generate our vesicles through emulsion transfer. That means we first need to generate water and oil emotions stabilized by lipids.
Because we want to study the exchange between two distinct populations of vesicles. We need two different kinds of emulsions. Here
we use two solutions with different fluorescently labeled DNA. In the next step,
we put these emulsions on top of a water-oil interface in our glass bottom microtiter plate.
Now we have a layer of oil which contains microdroplets on top of an aqueous layer beneath.
To get lipid vesicles from these droplets, we put the entire microtiter plate in a centrifuge and spin the droplets through the water-oil interface.
Here you can see a confocal microscope image of the liposomes we get from this process.
And now as I mentioned, we freeze and thaw these vesicles and as a result their contents mix.
Even though a similar behavior for the freezing and thawing of lipid vesicles has been shown before, our results differ quantitatively and qualitatively
from previous work as we point out in the paper. We further think the microtiter plate layout will enable efficient future screening experiments.
We looked into a number of different conditions; specifically
we varied freezing and thawing rate and surprisingly found that content mixing depends on both the freezing and the thawing rate.
We see the most efficient content mixing when we freeze our samples fast at minus 80 degrees Celsius and then slowly thaw them at room
temperature. In the paper you will find a detailed analysis of these experiments.
The biggest surprise was that the mechanism which we found responsible for this content exchange was
fundamentally different from previous reports for content exchange between vesicles. With these results
we hope to give a new insight in how basic environmental driving forces could have contributed to the emergence of life.

A man who previously claimed that he was injured when Dr. Phil hit him with his car might not have been entirely honest.
A new video obtained by TMZ shows the moment when the talk show host tried to pull in front of a van only to strike a man going by on his skateboard.
The man stands right up, takes off his headphones and briefly speaks with the driver, who the outlet reports was Dr. Phil. The two even shake hands before parting ways without further incident.
As E! News previously reported, the skateboarding man is named Terrence Bembury, who took to Instagram shortly after the accident in a series of photos that have since been deleted showing him in a hospital and alleging that he would have to miss two days of work because of his injuries.
He claimed he would seek legal action against Dr. Phil claiming he experienced pain and injuries that didn't manifest themselves until after the fact.
However, it seems clear from the video that it's unlikely the accident had any real lasting impact on him given his demeanor immediately following.
At the time, a spokesperson for Dr. Phil told the outlet that he talked to the man after the mishap and was assured he was OK. He said he'd cooperate with law enforcement if they had further questions.
After the release of the video, TMZ contacted Bembury's lawyer, John H. Mardoyan, who told the outlet that the video footage prompted him to rethink his decision to represent Bembury.
After viewing the video footage, we have decided to no longer represent Terrence Bembury in the matter, he said.

Hello everyone, as you can tell, this video is going to be in Spanish!
And yeah.. How are you? How is it going?
As some of you may know, a while ago
I started my YouTube channel and up to this video,
all of my videos were in English.
And I've got many comments and opinions
of people asking me
if I've forgotten how to speak Spanish,
if I only want to speak English or if I already consider myself Canadian.
And stuff like that.
So, even when I mentioned in my
first video why was I going to be uploading
videos in English
I thought it would be
a good idea to make a video in Spanish
since I can explain myself better in this language,
so it would be easier
for the people who watches this videos
and that speak Spanish,
to better understand
my motives. So in order for me
to better explain this
I need to tell you a little bit of me.
For all my life and until 8 or 9 months ago,
I've had always lived in Mexico City
so when I moved here to Canada
I had the opportunity to
be in touch with people from other countries and cultures,
something that had not been a possibility
for me in the past. And this is something
that I have enjoyed a lot and that I'm very interested in.
Besides from that I started to work here
and my work place happened to be in the touristic area
of the city in which I live, very close to the border.
Reason why we have a lot of tourists around.
And I personally have had the experience,
around three or four times,
of being treated very poorly by some of this people
after they've listened to the way I talk,
since my accent is very obvious
for people who natively speak English
and they can realize very easily that I speak Spanish, that I'm Latin.
So, as you can tell
this is a very non pleasant
experience for anyone.
After this happened,
I kept on thinking why did someone
who didn´t even know me, treat me like that.
Since they didn´t even knew my name
and actually I was
trying to be kind to them,
but they didn´t care and went on on being rude to me.
So then I realized that
definitively they were treating me like that
because all the things they´ve gathered and processed from the
latin or mexican culture.
They didn´t even give me and opportunity.
For them, that was the only way to treat people like me.
And I kept on thinking that even when there is mexican
or latin media that can reach people
all around the world,
to be honest,
there are only a few of them.
And the news, movies or series that
people from all around
can get access to
are not made my people from within our culture,
they've been interpreted and produced by external people.
So, someone else is telling our stories,
someone else is talking about
our ways of living,
someone else is deciding
how the world see us.
This to me, is something that keeps me from staying still,
something needs to be done.
So that's one of the reasons why I want to make this videos in English.
Because I don't see the point in talking about Mexico or Latin America
to Latin people.
We already know
the good and bad things we have.
What I want to do is to bring the opportunity
to other people in the world
of having a medium, even if it is very small
with which they can
obtain a different point of view,
a different perspective.
And of course, this tiny media is my YouTube Channel.
Besides Latinos, another reason for me
to keep going with this channel is
that there are other people, other communities and cultures, that suffer from the same pain.
Their stories are being told through
movies, series
and TV
but those really aren´t mediums that they create,
again they are being created by someone else.
So I think it´s valid
to try to talk about this other people
and let them tell their own story.
Hear it from them.
So this is why you'll see in my channel
somedays things
about Mexico, then something about Latinos,
about some other cultures,
and I'm planning on keeping to upload things about Canada,
because even when I've been treated poorly here in Canada
is always good to say
that it has never been by a Canadian, as far as I know.
Everyone has always treated me very nice here,
it's also a beautiful country
and one should always be grateful.
So yeah, that's why I'll
be posting about that too.
And that's a rough idea of what I want this channel to be.
I understand that there have been very few videos
and that it may be hard to get from them the whole idea
of this channel. So I ask for you to be patient,
to believe in me and
to accompany me in the next videos.
And well, I don´t know much about this yet,
I didn´t study communication
or acting or anything like that.
And I'm learning, I didn´t know how to properly use a camara
or to edit videos so I'm learning all along the way.
Because if there´s something about Mexican people is that nothing can stop us,
we are ingenious and you can do everything
with YouTube videos help.
So, those are the reasons
why I wanted to make this video so I could explain them,
So, all I ask is a little bit of your support
and
I hope to make of this channel something interesting,
something that you may like,
and even when the videos will be in English,
I´ll always put Spanish subtitles.
And, what else?
Oh yeah, I just
wanted to clarify that I love my country.
That I'm proudly Mexican,
that I speak Spanish and that Spanish to me is one of the most beautiful languages,
that it has a very big vocabulary,
and at least for me,
is the language in which I can express myself better.
You´ll realize, when I make videos in English
I have to record like 8 times and put together all the Dragon Balls
so I can get a decent(ish) video
And with this one I hope to record only once and do a little bit of editing.
